On a 2003-2006 Chevy 2500-3500 Express Van with a 4L80e transmission which trans would last longer given either a 4.8L engine or a 6.0L Engine?? and WHY??

Assume no towing and driving in the same conditions.

smaller motors reduce the amount of torque during full throttle upshifts...

full throttle upshifts are what wipes out transmissions...

ignoring transmission service needs also wipe out transmissions..  clogged filters reducing the ability of the pump to pull fluid up from the pan.. reducing the volume and pressure to create nice firm shifts..   reduced flows allow slippage between shifts and the clutches take it..  this causes additional heat which bakes the sealing rings.. shrinking them requiring more fluid flow...   its a downward spiral that ends deeply in your wallet..

if there was the ability to keep the transmission fluid at 165F.. the transmissions should last a LOT longer...
