The transmission on my girlfriends 2002 Buick Century (V6) intermittently hesitates after about 20 minutes of city driving.
I think a minor adjustment is necessary. She worries that a major repair is necessary.
Any hints as to how we can further troubleshoot this problem. Thanx in advance.

Check your Fluid Level and Color, Than Have it Scanned for any codes, a Local Auto Zone will do this for free

If by hesitation you mean that you have to rev the motor slightly and feel a slight bump to get the car moving after coming to a stop I would suspect a blown case cover gasket. Have seen this problem a few times before and there is a TSB about it that recommends dimpling the case and case cover in the problem area before installing the new gasket.
